薄层及凝胶色谱法测定树莓籽原花青素的平均聚合度

Determination of the mean Degree of Polymerization (mDP) from raspberry seeds by Thin Layer Chromatography (TLC) and Gel Chromatography (GPC)

  • 摘要: 在室温条件下,选择硅胶GF254为固定相,V(甲苯):V(丙酮):V(乙酸)=3:4:1为展开体系,15μL为点样量,通过薄层色谱法分离树莓籽原花青素得到三条谱带,由HPLC和凝胶色谱法分析测定,谱带Ⅰ为单体物质,谱带Ⅱ、Ⅲ平均聚合度为2.65、5.11。 

     

    Abstract: At room temperature, select silica gel GF254 as the stationary phase, V (toluene) :V (acetone) :V (acetic acid) =3:4:1 as the developing system, 15μL as the amount of sample application, separate raspberry seed proanthocyanidins by TLC and get three spectrums. HPLC was employed to analyze the compose and GPC was used to get the mDP from each spectrum of TLC. The composition of the spectrumⅠwas monomer material. The mDPs of spectrumⅡand Ⅲ were 2. 65 and 5. 11.
