嗜热真菌FL12产耐热木聚糖酶条件优化

Optimization of liquid culture conditions of a thermo-tolerant xylanase high-producing fungus FL12

  • 摘要: 自土样中筛选得到一株高产耐热木聚糖酶嗜热真菌FL12,通过单因素实验及Box-Behnkendesign响应面分析,确定该菌株液体发酵产耐热木聚糖酶最佳条件:碳源玉米芯粉(浓度为4.9%),氮源牛肉蛋白胨(1.0%),培养基初始pH6.45,装液量60mL,培养温度50℃,转速200r/min。在此条件下菌株FL12发酵7d产酶量达2284.06U/mL。结果显示该菌株发酵粗酶液最适温度75℃,85℃时相对酶活为62.94%,对高温耐受性较好。 

     

    Abstract: A thermophilic strain FL12 was isolated from soil samples, and its fermentation conditions of producing thermo-tolerant xyalnase were optimized by single-factor experiment and response surface analysis. The influences of medium composition and fermentation conditions on the fermentation were investigated. The optimal cultivation conditions were including carbon source was corncob at a concentration of 4. 9%, while the optimum nitrogen source was beef peptone (1. 0%) . A high xylanase activity of 2284. 06U/mL was obtained after 7d by incubation at 50℃ with initial pH6. 45, medium capacity of 60mL/250mL and rotation rate of 200r/min. The optimum temperature of FL12 crude fermentation broth was 75℃, 62. 94% residual enzyme activity was retained at 85℃.
