鼎湖鳞伞多糖的提取工艺及体内免疫调节活性的研究

Extraction and immunomodulatory activity of polysaccharide from Pholiota dinghuensis Bi

  • 摘要: 采用三因素三水平正交实验方法,研究了液料比、提取温度、提取时间这三个因素对鼎湖鳞伞多糖(Polysaccharide from Pholiota dinghuensis Bi,PFPD)提取工艺的影响。以雌性小白鼠作为模型实验,对其免疫器官指数、耳肿胀度、血清溶血素、血清和脾脏中溶菌酶的活性进行了研究。结果表明:PFPD最佳的提取工艺条件为:提取温度为90℃,提取时间为6h,液料比为20mL/g,PFPD的得率为8.95%。小鼠腹腔注射PFPD能使因环磷酰胺诱导产生的脾脏指数下降得以恢复,在一定程度上提高了机体的免疫力,三组不同剂量的PFPD可以显著地提高脾脏指数、迟发型超敏反应的右耳肿胀度、血清溶血素水平、脾脏和血清中溶菌酶的活性,且这种变化与鼎湖鳞伞粗多糖呈现剂量依赖关系。本实验表明鼎湖鳞伞多糖具有一定的体内免疫调节活性。 

     

    Abstract: A three - factor, three - level orthogonal array design was used to study the effects of three extraction conditions including temperature, time and solvent - to - solid ratio on polysaccharide yield. Intercellular polysaccharide was extracted from fermentation mycelium of Pholiota dinghuensis Bi. The activity of lysozyme in serum and spleen, immune organs index, swelling rate of ear and hemolysin in serum of mice were measured in vivo according to the animal models experiment.The results showed that the optimal conditions for the extraction of polysaccharide from Pholiota dinghuensis Bi ( PFPD) were 6h extractionat 90℃ and a solvent-to-solid radio 20mL /g, resulting in a polysaccharide yield of 8.95% . The PFPD intraperitoneal injected was able to overcome cyclophosphamide-induced immunosuppression.In addition, the spleen index, late onset allergic reaction of right ear degree, level of serum hemolysin and spleen, and serum lysozyme activity of the animals treated with different doses PFPD increased in a dose - dependent manner. The results suggested that the Pholiota dinghuensis Bi polysaccharide had some of immunomodulatory activity in vivo.
