安石榴苷的纯化及对金黄色葡萄球菌抑菌作用的研究

Purification of punicalagin and its antibacterial activity against Staphylococcus aureus

  • 摘要: 采用超声波辅助法从石榴皮中提取单宁类物质,并用Amberlite XAD-16大孔吸附树脂进行柱层析,以甲醇梯度洗脱法进行纯化,经高效液相色谱对其组分进行分析后,以安石榴苷含量最高的组分对金黄色葡萄球菌进行抑菌实验,分别测得抑菌圈直径和最小抑菌浓度,并检测了安石榴苷对金黄色葡萄球菌生长曲线的影响。液相结果表明,随着洗脱剂浓度的升高,安石榴苷的含量呈现先升高后降低的趋势,最高含量达79.6%;抑菌结果表明,安石榴苷纯化物对金黄色葡萄球菌有明显的抑菌效果,最小抑菌浓度为0.1mg/m L,安石榴苷纯化物以剂量依赖的形式抑制金黄色葡萄球菌的生长。 

     

    Abstract: Pomegranate peel tannins were extracted by ultrasonic-assisted extraction and purified by Amberlite XAD-16 macroporous resin. Gradient methanol was used as elution solution. Then the components were determined by high performance liquid chromatography (HPLC) analysis. The eluate with the highest content of punicalagin was used for the antimicrobial asssays. Inhibition zone diameters and the minimal inhibitory concentration (MIC) were investigated. The effect of punicalagin on the growth of Staphylococcus aureus was also monitored. HPLC showed that the purity of the eluate increased at first then decreased with the increasing concentration of ethanol. The highest purity among the different eluates was 79.6%. Antimicrobial study demonstrated that punicalagin exhibited good inhibitory effects against Staphylococcus aureus, the MIC was determined as 0.1mg/m L. Punicalagin inhibited the growth of Staphylococcus aureus in a dose-dependent manner.
