江苏东部沿海四种低值贝类软体中牛磺酸动态积累规律与适宜采收期研究

Study on the dynamic accumulation of taurine in four shellfish flesh of Jiangsu east costal and the appropriate harvest times

  • 摘要: 以采自江苏东部沿海的文蛤、青蛤、四角蛤蜊及菲律宾蛤仔四种低值贝类为样本,研究其软体中牛磺酸的动态变化规律,探讨这四种贝类的最适采收期。建立以邻苯二甲醛(OPA)柱前衍生化结合HPLC-UV测定牛磺酸的方法,以Kromasil 50mm×2.1mm C18为分离柱,甲醇-0.05mol/L乙酸钠(p H6.0)23∶77为流动相,流速0.3m L/min洗脱,紫外检测波长为330nm。该方法可以准确测定四种贝类软体中牛磺酸的含量,且灵敏度高,重现性好。贝类的生长具有明显季节性,一般春夏季较秋冬季生长较快,尤其夏季水温为2528℃时为快速生长期,此时,贝类软体中牛磺酸含量达到最高。菲律宾蛤仔牛磺酸含量最高在7月,约为42.79mg/g;而文蛤、青蛤、四角蛤蜊软体中牛磺酸含量最高在6月,分别约为27.48、16.59、15.00mg/g。若从产量和有效成分牛磺酸的含量变化上综合考虑,建议菲律宾蛤仔牛磺酸最佳采收期为7月,文蛤、青蛤、四角蛤蜊最佳采收期为6月。 

     

    Abstract: Four kinds of shellfish from the east coast of Jiangsu province were collected to research the dynamic changes of taurine in the shellfish flesh,in order to investigate the appropriate harvest times. To determine the content of taurine,a method of HPLC- UV combined with opthaldialdehyde was established,and the samples extracted by 70% alcohol were separated by a Kromasil 50 mm × 2.1mm C18 column with the column temperature of40℃.The mobile phase was composed of 23% methanol and 77% 0.05 mol / L sodium acetate( p H6.0) at the flow rate of 0.3m L / min with gradient elution.The UV detection wavelength was 330 nm.This method,with high sensitivity,good reproducibility,could accurately determine the content of taurine in four shellfish. Generally,the growth of shellfish had an obvious seasonal characteristic and its growth faster in spring and summer than autumn and winter,especially when water temperature was 25 to 28℃ in summer- the rapid growth period. During the period,the content of taurine reached its peak.Moreover,the content of taurine of Ruditapes philippinarum reached highest was July,with 42.79 mg / g. Meretrix meretrix Linnaeus,Cyclina sinensis,Mactra veneriformis Reeve was in June,when the content of taurine was up to 27.48,16.59,15.00 mg / g,respectively.In combination production with content changes of effective constituent- taurine,the best harvest time of Ruditapes philippinarum was recommended in July,with Meretrix meretrix Linnaeus,Cyclina sinensis,Mactra veneriformis Reeve was in June.
