响应面法优化碱性蛋白酶提取瘤背石磺肌肉蛋白

Optimizing conditions for alkaline protease extraction of Onchidium struma mantle protein by response surface methodology

  • 摘要: 以瘤背石磺肌肉为原料,蛋白提取率为指标,采用碱性蛋白酶提取瘤背石磺肌肉中的蛋白。在单因素实验的基础上,通过响应面法考察温度、p H、时间和液固比对蛋白提取率的影响,优化碱性蛋白酶酶解反应的工艺条件。结果表明,响应面法得到的最佳工艺条件合理可行,最佳工艺条件为温度56℃,p H12.6,时间3.2 h,液固比93 m L/g。在上述条件下,蛋白提取率的预测值为18.36%,实际值为18.30%,两者相对误差0.34%,表明此模型合理可行。 

     

    Abstract: The effect of alkaline protease on the rate of protein extraction of Onchidium struma mantle were studied.Then the enzymolysis conditions of alkaline protease were optimized by using response surface method(RSM)on the basis of single factor as temperature,p H value,time,liquid-solid rate and enzyme amount experiments.The results showed that the optimum enzymolysis conditions were optimized as following:temperature 56 ℃,p H value 12.6,time 3.2 h,liquid-solid rate 93 m L/g respectively.Under these conditions,the predicted protein extraction rate was 18.36%,compared to the measured value 18.30%,the relative error was 0.34%. Results showed that the conditions were reasonable and feasible.
