贯叶连翘花中挥发油的提取工艺优选及抑菌活性研究

Extraction and antibacterial activity of volatile oil from Hypericum perforatum L.flowers

  • 摘要: 采用水蒸气蒸馏法对贯叶连翘花中挥发油进行提取,以挥发油提取率为响应值,通过单因素实验对影响提取工艺的参数(浸泡时间、提取时间和料液比)进行考察,并利用三因素三水平响应曲面实验法确定了贯叶连翘花中挥发油的最优提取工艺,即浸泡时间19 h,液固比9∶1 m L/g,提取时间10 h,该条件下贯叶连翘花中挥发油的提取率为1.03 m L/100 g,与理论值1.10 m L/100 g仅相差2.73%。同时测试了该挥发油的抑菌活性,其对大肠杆菌、金黄色葡萄球菌和枯草芽孢杆菌的抑菌圈直径分别为9、10、12 mm,表明贯叶连翘花中挥发油有一定的抑菌活性。 

     

    Abstract: To study steam distillation extraction technology of volatile oil from Hypericum perforatum L. flowers,the type of parameters soaking time,solvent to solid sample ratio and extraction time were investigated in single factor experiments with extraction yield of volatile oil as evaluation index.The best extraction processes were obtained with three factors and three levels of response surface methodology as follows: soaking time 19 h,solvent to solid sample ratio 9∶ 1 m L / g and extraction time 10 h respectively,leading to an extraction yield of volatile oil 1.03 m L /100 g,and the experimental value are basically close to the theoretical expectation value 1.10 m L /100 g,with only 2.73% deviation. Combining with the antibacterial activity tests,the antimicrobial cycle diameter of volatile oil from Hypericum perforatum L. flowers were 9,10,12 mm for Escherichia coli,Staphyloccocus aureus and Bacillus subtilis,which suggested volatile oil from Hypericum perforatum L. flowers has a certain antibacterial effect.
