5’-核苷酸对慢性酒精中毒诱导大鼠结肠功能损伤的影响

Effects of 5'-nucleotides on the injury of colon function induced by chronic alcoholism in rats

  • 摘要: 探讨外源性5’-核苷酸对酒精诱导大鼠结肠功能紊乱的保护作用。50只SPF级雄性Wistar大鼠随机分为正常对照组、酒精模型组、等热量对照组及0.04%、0.16%核苷酸组。连续酒精灌胃6周后,结果发现,外源性5’-核苷酸能够有效改善酒精引起的大鼠结肠病理性改变,显著提高结肠功能相关蛋白Claudin和Occludin的表达水平(p<0.01或p<0.05),极显著降低大鼠结肠组织中Toll样受体4(TLR4)和分化抗原簇14(CD14)的表达水平(p<0.01);外源性5’-核苷酸能够显著增加大鼠回肠内容物中双歧杆菌(酒精组8.00 log CFU/g,核苷酸组8.32 log CFU/g)和乳酸杆菌数量(酒精组7.44 log CFU/g,核苷酸组8.13 log CFU/g)(p<0.05),同时显著减少肠球菌(酒精组7.52 log CFU/g,核苷酸组7.03 log CFU/g)及大肠杆菌数量(酒精组6.85 log CFU/g,核苷酸组6.19 log CFU/g)(p<0.05)。因此,外源性5’-核苷酸能够通过调节肠道菌群改善酒精引起的大鼠结肠损伤。 

     

    Abstract: The aim of this study was to investigate the protective effects of exogenous 5'-nucleotides on alcohol induced colon function injury in SD rats.Fifty SPF male Wistar rats were randomly divided into normal control group, alcohol model group, dextrose control group, 0.04% and 0.16% nucleotides intervention group. After continuous ethanol lavage for 6 weeks, the results showed that exogenous 5'-nucleotides could effectively improve the pathological changes of colon in rats induced by alcohol, increase the expression levels of Claudin and Occludin and reduce the levels of TLR4 and CD14 in colon tissues ( p <0.01) .The numbers of Bifidobacterium ( 8.32 vs.8.00 log CFU/g) and Lactobacillus ( 8.13 vs.7.44 log CFU/g) in ileum contents were significantly decreased, and the numbers of Enterococcus ( 7.03 vs.7.52 log CFU/g) and E. coli ( 6.19 vs.6.85 log CFU/g) were significantly decreased in nucleotides intervention groups when compared with the alcohol model group ( p < 0.05) .Therefore, exogenous 5'-nucleotides could improve the alcohol induced colon function injury in rats which may attribute to the regulation of gut microbiota.
