牦牛血胰蛋白酶水解液中抗菌肽的筛选研究

Study on identification of antimicrobial peptides from trypsin hydrolysate of yak blood

  • 摘要: 以牦牛血的胰蛋白酶水解液为原料,用Sephadex G-15凝胶柱,在以10 mmol/L磷酸盐缓冲液(pH7.0)为洗脱液,流速为0.4 mL/min的洗脱条件下,利用抗菌肽与细菌细胞膜的吸附结合特性,分离出两个含抗菌肽组分的洗脱峰。研究显示抑菌活性大的洗脱峰再经过RP-HPLC反复纯化,收集组分并检测抑菌活性,得到两个色谱纯的抗菌肽YakB-1和YakB-2。抑菌实验表明,抗菌肽YakB-1/2对细菌和真菌都具有广谱抗菌活性;抗菌肽YakB-2无论对G-菌还是G+菌的抗菌活性都比YakB-1强,但YakB-1对真菌的抑菌活性更强。研究结果可为牦牛血资源开发与利用提供参考依据。

     

    Abstract: Two eluting peaks including antimicrobial peptide component were isolated from trypsin zymolyte of yak blood, using the membrane binding property of antimicrobial peptides with bacteria cell, with Sephadex G-15 column, under conditions of 10 mmol/L phosphate buffer as eluent and flow rate 0.4 mL/min. The results of binding assay showed that the two novel antimicrobial peptides YakB-1 and YakB-2 were successfully isolated, combining the repeated purification with reverse phase high-performance liquid chromatography followed by antimicrobial activities assay. Antimicrobial activities indicated that both peptides clearly displayed a very impressive broad spectrum of activities against bacterial and fungus tested. Furthermore, the antimicrobial activities of YakB-2 were more powerful than that of YakB-1 against Gram-positive and Gram-negative bacterial. However, YakB-1 showed stronger antimicrobial activities against fungus than YakB-2.The results provide references for the development and utilization of yak blood resource.
