牛骨胶原低聚肽促进皮肤切口手术模型小鼠术后伤口愈合的机制研究

The Mechanism of Bovine Collagen Oligopeptide Promoting Wound Healing in Mice after Skin Incision Surgery

  • 摘要: 探讨牛骨胶原低聚肽促进皮肤切口手术模型小鼠术后伤口愈合的可能机制。96只SPF级ICR雄性小鼠随机分为空白对照组和3个牛骨胶原低聚肽剂量组(低剂量、中剂量、高剂量,灌胃剂量分别为0.375、0.75、1.5 g/kg·bw),每组24只。建立小鼠背部皮肤切口手术模型,术后连续灌胃11 d,并在术后第3、7、11 d分批处死8只小鼠进行相关指标的检测。结果发现,术后第11 d,牛骨胶原低聚肽高剂量组小鼠抗张力强度显著高于空白对照组(p<0.05);术后第3、7和11 d,低剂量、中剂量和高剂量牛骨胶原低聚肽能够显著增加小鼠血清中白蛋白和前白蛋白浓度(p<0.05);术后第3、7和11 d,中剂量和高剂量牛骨胶原低聚肽能够显著促进血清白细胞介素-10的表达水平(p<0.05),术后第7、11 d,牛骨胶原低聚肽能够显著抑制肿瘤坏死因子-α的表达水平(p<0.05)。因此,牛骨胶原低聚肽可以有效促进小鼠伤口愈合,其作用机制可能与其提高小鼠营养状况及抑制炎症反应有关。

     

    Abstract: In order to investigate the mechanism of bvine collagen oligopeptide(BCOPs)promoting wound healing in mice after skin incision surgery. A total of 96 SPF ICR male mice were randomly divided into control group and 3 BCOPs intervention groups(0.375,0.75,1.5 g/kg·bw,respectively),and 24 mice in each group. The mice were carried out by mice stomach-lavaging continuously for 11 d after incision wound model. 8 mice each group were sacrificed separately at 3rd d,7th d,and 11th d. It was found that the tension strength of mice in the high-dose BCOPs group was significantly higher than that in control group at day 11.Low,medium and high dose BCOPs could significantly increase the concentrations of ALB and PA in serum at 3rd d,7th d,and 11th d(p<0.05). The medium and high dose BCOPs could markedly promote the expression levels of IL-10 in serum at day 3,7 and 11(p<0.05),while BCOPs could also significantly inhibit the expression levels of TNF-α(p<0.05). Therefore,BCOPs can effectively promote the wound healing in mice,which may be related to improving the nutritional status and inhibiting the inflammatory response in mice.
