西番莲果皮多糖微波辅助提取工艺优化及其体外抗氧化性

Optimization of Microwave-assisted Extraction Process of Polysaccharide from Passiflora edulis Sims Peel and Evaluation of Antioxidant Activity in vitro

  • 摘要: 利用响应面优化微波辅助提取西番莲果皮多糖工艺,并研究其体外抗氧化活性。在单因素实验的基础上,根据Box-Behnken实验设计对料液比、提取时间和微波功率条件对多糖提取率的影响进行优化和分析。确定微波辅助提取最佳工艺参数:料液比1:27 g/mL,提取时间3.4 min,微波功率420 W,此条件下提取率为14.12%±0.41%,是传统水浴提取的1.5倍。西番莲果皮多糖体外抗氧化实验表明:微波辅助提取的西番莲果皮多糖在浓度为1.0 mg/mL时,DPPH·和·OH的清除率分别为74.02%和14.41%,其IC50值分别为0.374和61.06 mg/mL。

     

    Abstract: Response surface methodology was applied to optimize the microwave-assisted extraction process of polysaccharide from Passiflora edulis Sims peel and its antioxidant activities in vitro were evaluated. On the basis of single factor test,the ratio of raw material weight to water volume,the extraction time,the microwave power were analyzed and optimized by the Box-Behnken design. The optimized parameters of the microwave-assisted extraction technology were obtained as follows:the ratio of raw material weight to water volume was 1∶27;the extraction time was 3.4 min;the microwave power was 420 W and the extraction rate of polysaccharide was 14.12%±0.41%,which was 1.5 times of that of traditional water bath extraction. Antioxidant activities in vitro of polysaccharide indicated that the DPPH· and ·OH scavenging rates of polysaccharide were 74.02% and 14.41% respectively when the concentration was at 1.0 mg/mL,and the IC50values of 0.374 and 61.06 mg/mL,respectively.
