非标记定量蛋白质组方法分析鲟鱼肽抗D-半乳糖导致的小鼠衰老作用的研究

摘要: 为了研究鲟鱼肽抗衰老作用机制,利用非标记定量蛋白质组方法对去除高丰度蛋白后的灌胃鲟鱼肽小鼠血清进行蛋白质组分析。分析结果表明,从去除高丰度蛋白后的血清中共获得442种蛋白,其参与21个生理过程,具有7类分子功能,存在于10个细胞组分中。将模型组与治疗组进行t-test分析,得到3个差异表达蛋白质。在进行生物信息学分析时,通过扩大筛选范围,又增加了6种蛋白质,其中二磷酸甘油酸变位酶(AC:P15327)、载脂蛋白D(AC:P51910)、黄素还原酶(AC:Q923D2)和过氧化物酶-2(AC:Q61171)的上调以及巯基氧化酶1(AC:Q8BND5)的下调可以提高机体清除自由基的能力。这些数据表明,鲟鱼肽抗衰老能力是通过提高机体清除自由基的作用来发挥的。Abstract: To investigate anti-aging mechanism of A. schrenki Brandt peptides (ASP), label-free quantitative proteomics was used. The results of LFQ (Label Free Quantization) showed 442 low-abundant proteins in serum in mice treated with ASP. These proteins covered 21 biological processes, 7 molecular functions and 10 cellular components. There were 3 differentially expressed proteins according to t-test. In bioinformatics analysis, the screening criteria were expanded to 9 proteins. Among the 9 proteins, the up-regulated bisphosphoglycerate mutase (AC:P15327), apolipoprotein D (AC:P51910), flavin reductase (AC:Q923D2) and peroxiredoxin-2 (AC:Q61171) and down-regulated sulfhydryl oxidase 1, (AC:Q8BND5) could improve the ability to scavenge free radicals. These results showed that the ASP prevented aging by scavenging free radicals.
