肉苁蓉总多酚纯化工艺及其抗运动性疲劳作用研究

Purification and Anti-motor Fatigue Effect of Total Polyphenols from Cistanche Extraction

  • 摘要: 为优化大孔树脂纯化肉苁蓉多酚提取物的工艺条件,通过静态吸附与洗脱试验筛选合适的大孔树脂型号,并研究其吸附等温与吸附动力学模型后,采取动态吸附与洗脱单因素实验确定最佳纯化工艺条件,同时进行动物的抗运动性疲劳研究。实验结果表明,HPD-400大孔树脂的吸附等温方程符合Langmuir模型,吸附量随温度升高而降低,且符合准二级动力学吸附过程。采用大孔树脂纯化肉苁蓉多酚提取物的最佳工艺条件为:配制体积60 mL,6 mg/mL上样溶液,以2 mL/min流速上样至HPD-400大孔树脂饱和吸附后,采用体积为180 mL,60%乙醇溶液,以1 mL/min流速洗脱,提取物中肉苁蓉多酚纯度由20.12%提高至42.63%。与空白对照组相比,不同剂量的肉苁蓉多酚纯化产物可显著延长小鼠游泳力竭时间(P<0.05,P<0.01),显著增加体内肝糖原、肌糖原含量与乳酸脱氢酶活力(P<0.05,P<0.01),并有极显著降低乳酸浓度水平(P<0.01),因此可较好地缓解机体疲劳,从而为肉苁蓉多酚化合物的后续开发利用提供参考。

     

    Abstract: To optimize the purification process condition of total polyphenols of extract from Cistanche by macroporous resin, the HPD-400 macroporous resin type was chosen through static adsorption-desorption experiments, meanwhile, isothermal adsorption model and adsorption kinetics model of the resin were furthermore investigated and then anti-motor fatigue of animal experiment was carried out. The results showed that equilibrium adsorption data of HPD-400 macroporous resin for total polyphenols were fitted Langmuir isotherm equation, the adsorption decrease with the increasing of temperature, and the kinetics fitted the quasi-second-order kinetics equation. The optimum purification process conditions were as follows:the initial concentration in sample solution 6 mg/mL, flow rate 2 mL/min, loading sample volume 60 mL, and 60% (v/v) ethanol 180 mL was used as eluent with flow rate of 1 mL/min. After purification, the purity of total polyphenols in product was increased from 20.12% to 42.63%. Compared with the blank control group, the groups of polyphenols of Cistanche at all doses significantly prolonged weight-loaded swimming time of mices (P<0.05, P<0.01), obviously improved activity of lactic dehydrogenase and storage of hepatic glycogen and muscle glycogen in vivo (P<0.05, P<0.01), decreased lactic acid level after sporting (P<0.01).Therefore, polyphenols of Cistanche could enhanced sporting stamina and had good resisting movement fatigue effect, the study could provide reference for its subsequent development and exploitation.
