河豚鱼皮胶原寡肽螯合锌的体内体外抗氧化活性研究

Study on In Vivo and In Vitro Antioxidant Activity of Collagen Oligopeptide Chelated Zinc from Puffer Skin

  • 摘要: 以河豚鱼皮为原料,制备了河豚鱼皮胶原寡肽螯合锌,并对螯合锌的基本组成成分和结构进行了表征。以1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、羟基自由基(·OH)的清除能力、三价铁(Fe3+)的还原能力为指标,考察河豚鱼皮胶原寡肽螯合锌的体外抗氧化能力,实验表明,河豚鱼皮胶原寡肽螯合锌对DPPH自由基、羟自由基的清除率IC50值分别为2.54和7.01 mg/mL,并且具有较强的三价铁还原能力,同时河豚鱼皮胶原寡肽螯合锌的抗氧化活性明显优于河豚鱼皮胶原寡肽。体内抗氧化活性实验结果显示,河豚鱼皮胶原寡肽螯合锌高剂量组与缺锌组比较能显著提高大鼠血清中总抗氧化能力(T-AOC)、谷胱甘肽(GSH)和总超氧化物歧化酶(T-SOD)的活力,而使丙二醛(MDA)的含量明显降低(P<0.05,0.01),并且其抗氧化活性要优于同剂量的葡萄糖酸锌组。河豚鱼皮胶原寡肽螯合锌具有良好的体内体外抗氧化活性,有望作为新型抗氧化剂应用于生物制品和食品工业中。

     

    Abstract: Collagen oligopeptide chelated zinc was prepared from puffer skin. Its composition and structure were characterized. Taking 1,1-diphenyl-2-trinitrobenzene hydrazine free radical(DPPH·),hydroxyl free radical(·OH)scavenging ability,ferric iron(Fe3+)reducing power as indicators,the in vitro antioxidant capacity of collagen oligopeptide chelated zinc was evaluated. The experimental results showed that both collagen peptide and zinc chelate had DPPH free radical and hydroxyl free radicals scavenging activities,and ferric iron(Fe3+)reducing power. They also had good dose-effect relationship with the sample concentration. Collagen oligopeptide chelated zinc showed greater antioxidant ability than collagen oligopeptide. Collagen oligopeptide chelated zinc could significantly improve the total antioxidant capacity(T-AOC),glutathione(GSH)and total superoxide dismutase(T-SOD)activity in the serum of Zn-deficient rats,and obviously reduce the content of malondialdehyde(MDA)(P<0.05,0.01). Collagen oligopeptide chelated zinc from puffer skin has excellent antioxidant activity in vitro and in vivo,which is expected to be used as a new antioxidant in biological products and food industry.
