Determination of Isomaltooligosaccharide in Foods by Ion Chromatography-pulsed Amperometric Detection

  • Objective: An ion chromatography - pulsed amperometric detection method was developed for the determination of isomaltose, isomaltotriose, panose in foods. Methods: Samples were extracted by water and the protein in sample was deposited by acid, and purified with the column of guard ⅡRP, then those sugars were separated on a CarboPac PA20 high performance anion-exchange column of ThermoFisher. The water(A), 200 mmol/L sodium hydroxide solution (B) were used as mobile phases for gradient elution, and pulsed amperometric detector was used for quantitative determination. Results: The linear relationship was good within the range of 0.8~16.0 mg/L, the correlation coefficient was more than 0.999, the RSD of precision of isomaltose、isomaltotriose and panose were 3.42%, 2.34%, 1.11%, the recovery rate was 85.63%~105.22%, the RSD was 2.48%~4.49%. Conclusion:This method demonstrated to be simple with good accuracy and specifity, which is suitable for the determination of isomaltooligosaccharide in kinds of foods.
