HPLC method development for analysis of phenolic acids and flavone C- glycosides in Bambusa textiles McClure

  • An analytical method based on high performance liquid chromatography( HPLC) was developed for quantification of three phenolic acids and four flavone C- glycosides in Bambusa textiles Mc Clure,and was also validated.The optimum chromatographic conditions were determined using a C18column( 4.6 mm × 150 mm,5 μm)at the detection wavelength of 254 nm and a flow rate of 1.0 m L / min.Mobile phase was a binary eluent of methanol( A) and 0.1% aqueous phosphoric acid( v /v)( B) with gradient conditions as follows: 0 ~ 10 min,10% ~ 22% A,10~30 min,22% ~35% A,30 ~ 40 min,35% ~ 60% A. As a result,p- hydroxybenzoic acid,vanillic acid,p- coumaric acid,orientin,isoorientin,vitexin and isovitexin had good linearity( R2≥0.9996) within range of 0.08~100,0.04~100,0.25~50,0.24~60,0.22~56,0.06 ~ 56 and 0.12 ~ 60 μg / m L,respectively. The recoveries of the analytes were between96% to 103%( RSD < 3%),and the RSD of intraday and interday precisions were all less than 3.48%. The method was of high sensitivity,good reproducibility,and meets the methodological requirements. The method could apply to determine the contents of three phenolic acids and four flavone C- glycosides in B.textiles.
