响应面法优化枯草芽孢杆菌产胞外多糖培养基

Optimization of fermentation culture medium of extracellular polysaccharide from Bacillus subtilis by response surface methodology

  • 摘要: 从酒药中分离筛选出一株产胞外多糖枯草芽孢杆菌(Bacillussp.)。以胞外多糖的产量为指标,通过响应分析法对细菌胞外多糖产生的发酵培养基进行了初步研究。首先利用单因子实验对培养基中不同成分及添加量进行优化,然后在此基础上利用Box-Behnken中心组合实验设计对影响胞外多糖产量的因素进行优化分析,从而获得最适产胞外多糖的培养基组成。研究结果表明,枯草芽孢杆菌产胞外多糖最佳培养基组成为(g/L):蔗糖25.449、蛋白胨15.229、柠檬酸三钠2.971、牛肉膏3.0、硫酸镁1.0,在此条件下枯草芽孢杆菌胞外多糖的产量为3.554g/L。 

     

    Abstract: Extracellular polysaccharide by Bacillus subtilis strain used in this study was isolated from Chinese yeast.The optimum culture medium for exopolysaccharide production by a Bacillus subtilis strain were primarily studied in the response surface method.First, optimum culture medium and additive amount were adopted by single factor experiments.On the basis of single factor experiment, the best carbon source, nitrogen source and inorganic salt were acquired by response analysis in which the exopolysaccharide production were the main indicators.The results indicated that the optimum composition medium was as follows:sucrose 25.449g/L, peptone bacto 15.229g/L, trisodium citrate dihyhrate 2.971g/L, beef extract 3.0g/L, magnesium sulfate 1.0g/L.Under the conditions, the production of extracellular polysaccharide by Bacillus subtilis was 3.554g/L.
