烟草中果糖嗪的超声提取工艺研究

  • 摘要: 建立了烟草中果糖嗪的超声提取工艺和高效液相色谱含量测定方法。提取工艺为:萃取剂为纯水,萃取时间40min,液固比50:3(mL/g)。色谱条件为:分离柱为Waters Carbohydrate Analysis柱(3.9mm×300mm),流动相为乙腈:水(V/V)=80:20,流量1.0mL/min,柱温为室温,检测波长275nm,进样体积10μL。结果:标准曲线的相关系数的平方为0.9969,线性范围6~14μg/mL,果糖嗪的平均回收率为98.7%,相对标准偏差RSD均小于3%,检测限为0.5μg/mL,烟草中果糖嗪的质量百分含量约为0.02%,适用于烟草样品中果糖嗪的定量分析。 

     

    Abstract: Fructosazine content in tobacco with ultrasonic extraction-high performance liquid chromatography method was established.Extract processes were as follows:extraction agent for pure water, extraction time 40min, liguid solid ratio 50:3 (mL/g) .Chromatographic conditions were separation column Waters Carbohydrate Analysis (3.9mm×300mm) , mobile phase acetonitrile:water (V/V) 80:20, flow velocity 1.0mL/min, room column temperature, wavelength detection 275nm, sample volume 10μL.The results showed that the correlation coefficient of standard curve was 0.9969, the linear ranged of 6 ~ 14μg/mL, fructose in the average rate of recovery was 98.7%, the relative standard deviation was less than 3%, the detection limit was 0.5μg/mL in tobacco.Fructosazine in the tobacco was about 0.02% in the mass percent concentration.This method was suit for analyzing the content of fructosazine to tobacco samples in quantitative.
