金橙黄微小杆菌产碱性蛋白酶培养基组成及发酵条件优化

Optimization of alkaline protease-producing Exiguobacterium aurantiacum on medium compositions and fermentation process

  • 摘要: 采用单因素实验和正交实验的方法,对金橙黄微小杆菌产碱性蛋白酶条件进行优化。结果表明,最佳发酵培养基组成成分为酵母膏10g/L、玉米粉15g/L、NaCl15g/L,摇瓶培养最佳条件为初始pH8.8,培养时间28h,温度25℃,接种量6%条件下,在此条件下酶活性达到37.793U/mL。金橙黄微小杆菌对生长条件要求不高、容易培养,碱性蛋白酶产量较高,表现出了潜在的工业开发价值。 

     

    Abstract: The culture medium composition and fermentation conditions for protease producing from Exiguobacterium aurantiacum had been studied with univariate tests and orthogonal experiment in this paper.The optimal medium compositions was as follows 10g/L yeast extract, 15g/L corn starch and 15g/L NaCl, and the best fermentation conditions was as initial pH8.8, time for 24h, temperature for 25℃, inoculum 6%.The protease activity reached 37.793U/mL.E.aurantiacum could grow in poor condition, and the alkaline protease production was relatively high.The E.aurantiacum had potential to be used to produce alkaline protease.
