直投式青海弧菌冻干粉保护剂的筛选及优化

Screening and optimization of the freeze-dried protectants for the lyophilized Vibrio-qinghaiensis of direct vat set

  • 摘要: 为了得到直投式青海弧菌Q67在冷冻干燥过程中最好的保护剂,首先单因素筛选出效果较好的保护剂,再采用响应面分析法对青海弧菌Q67发光菌冻干保护剂进行优化研究。结果表明:脱脂乳、蔗糖、可溶性淀粉、谷氨酸钠对菌种Q67保护作用突出,以冻干后菌粉复苏30min的相对发光率(RLU)为响应值,当以脱脂乳7.68%、蔗糖2%、可溶性淀粉0.25%、谷氨酸钠1%作为混合保护剂使用时,能使青海弧菌的相对发光率高达1382.87%。并当保护剂:菌体=1:1和作用时间为30min时,青海弧菌的相对发光率高达1356%。 

     

    Abstract: In order to obtain the best protectants for Vibrioqinghaiensis sp.-Q67 of direct vat set, the freeze-dried protectants was firstly screened by single factor and then optimized by response surface analysis.The study indicated that in many substances the skim milk, saccharose, soluble starch and sodium glutamate have outstanding protective effect.In response to the relative light unit (RLU) of the lyophilized Vibrioqinghaiensis sp.-Q67 after the regeneration for 30min, when taken skim milk 7.68%, sucrose 2%, soluble starch 0.25%, sodium glutamate 1% as the combined optimal formula, the relative light unit (RLU) could reach 1382.87%;when the volume of protectants was equal to the strain and the response time was 30min, the relative light unit (RLU) could achieve 1356%.
