响应面分析法优化1-脱氧野尻霉素液相色谱检测的衍生化条件

Optimization of derivatization conditions for the determination of 1-deoxynojirimycin by response surface methodology

  • 摘要: 在1-脱氧野尻霉素的高效液相色谱法分析中,利用响应面分析法优化了荧光衍生化的最佳条件。根据Box-Behnken中心组合实验设计原理,在单因素实验基础上,以1-脱氧野尻霉素回收率为响应值,应用响应面分析法研究衍生化反应时间、反应温度和衍生化试剂用量各因素间的交互作用及对1-脱氧野尻霉素回收率的影响。确定最佳衍生化条件为:衍生化反应时间22min、衍生化反应温度29℃、衍生化试剂用量0.43mmol/L,实际回收率为99.4%。 

     

    Abstract: For the purpose of establishing a high performance liquid chromatography method for the determination of 1-deoxynojirimycin (1-DNJ) , the optimal derivatization conditions were studied by response surface methodology. Based on the result of single factor experiments, the Box-Behnken design was used to investigate the effect of derivative time, temperature and the amount of derivative reagent on the recovery of 1-deoxynojirimycin. The derivative conditions were found to be 22min, 29℃ and 0.43mmol/L and the recovery was 99.4%.
