赤丹皮中芍药苷的不同提取工艺研究

Research of different extraction process of paeoniflorin from root bark of Paeonia delavayi var.lutea

  • 摘要: 对赤丹皮中芍药苷进行提取工艺的考察。采用水浴法、回流提取和超声提取后经HPLC进行含量测定。利用正交分析,得到3种提取工艺的最优条件为:水浴法:50%乙醇,提取1h,料液比为1:450,提取2次,最佳工艺条件提取芍药苷提取含量为27.217mg/g;回流提取法:50%乙醇,提取时间2h,料液比1:450,提取3次,采用其最佳工艺条件提取芍药苷提取含量为26.688mg/g;超声提取法:90%乙醇,提取时间30min,料液比1:450,提取3次,采用其最佳工艺条件提取芍药苷提取含量为30.468mg/g。建立的方法简便、可靠,重复性好,可为赤丹皮的全面研究提供一定的数据参数。 

     

    Abstract: To study the extraction process of paeoniflorin in root bark of Paeonia delavayi var.lutea.The paeoniflorin was extracted by water, reflux extraction and ultrasonic extraction and the content was assayed by HPLC.Using orthogonal experiments analysis, the optimal extraction conditions of three extraction process were as follows:the water bath extraction method:50% ethanol, extraction time 2h, ratio of material to solvent 1∶450, extract 2 times.Content of the optimum conditions of extraction of paeoniflorin was 27.217mg/g.The reflux extraction method:50% ethanol, extraction time 2h, ratio of material to solvent 1∶450, extract 3 times.Content of the optimum conditions of extraction of paeoniflorin was 26.688mg/g.The ultrasonic cell crushing extraction method:90% ethanol, extraction time 30min, ratio of material to solvent 1∶450, extract 3 times.Content of the optimum conditions of extraction of paeoniflorin was 30.468mg/g.Methods established were simple, reliable and reproducible, which could provide data basis in a comprehensive study of root bark of P.delavayi var.lutea.
