野生牛肝菌多糖提取工艺的优化及其对自由基的清除作用

Optimization of extraction technology and scavenging activity to free radicals of polysaccharides from Natural Boletus

  • 摘要: 以多糖产率为指标,在单因素实验基础上,选用正交实验对野生牛肝菌粗多糖的提取工艺进行优化,再经乙醇沉淀、脱蛋白和真空干燥后得到野生牛肝菌多糖。结果表明,野生牛肝菌粗多糖最佳提取工艺为温度为75℃,浸提时间为2h,在料液比1:25下提取3次,多糖产率为9.13%。影响野生牛肝菌粗多糖提取的主要因素是料液比和提取温度,其次是提取次数。应用化学发光法对野生牛肝菌粗多糖的.OH和O-2.的清除能力进行了研究,结果表明,野生牛肝菌粗多糖对.OH和O-.2自由基均具有明显的清除能力,清除O-2.自由基的IC50为5.40mg/mL,清除.OH自由基的IC50为6.36mg/mL,野生牛肝菌粗多糖对O-.2自由基的清除能力是对.OH自由基的清除能力的1.18倍。 

     

    Abstract: On the basis of single factor test, optimization of extraction process of crude Natural Boletus polysaccharides was investigated by orthogonal tests.Then the Natural Boletus polysaccharide was obtained after ethanol-precipitation, deproteinixation and vacuum drying.The results showed that the optimum conditions for the isolation was as follows:temperature was 75℃, time was 2h, material to water ratio was 1∶25, times was 3, polysaccharide yield was 9.13%.The major factors affecting Natural Boletus polysaccharides extraction were material to water ratio and temperature in the first place, and times in the next place.The scavenging ability of ·OH and O-2· of Natural Boletus polysaccharide were assayed by chemiluminescence.There results showed that Natural Boletus polysaccharide had evident abilities of scavenging hydroxyl radicals and superoxide anion radicals.The crude Natural Boletus polysaccharides obtained had strong superoxide anion radicals scavenging effect, with an IC50 of 5.40mg/mL, which was more than 1.18-fold increased after hydroxyl radicals, revealing an IC50 of 6.36mg/mL.
