滑子菇多糖超声辅助提取工艺及抗氧化活性评价

Ultrasonic synergistic extraction and antioxidation of polysaccharides from Pholiota nameko

  • 摘要: 以滑子菇为原料,通过正交实验获得滑子菇多糖的超声波辅助提取最佳工艺,并对清除DPPH·、O2-·和·OH和还原能力进行测定。超声处理后,采用水提取法,并用苯酚-硫酸法测定滑子菇多糖中多糖的得率。通过单因素实验和正交实验考察超声时间、超声功率以及液料比对滑子菇多糖得率的影响,确定超声频率为80Hz、超声时间为15min以及液料比为20∶1是最佳工艺,多糖得率为11.219%。通过对清除自由基和还原能力评价,结果显示滑子菇多糖具有一定的抗氧化活性。 

     

    Abstract: The orthogonal experiment was used to optimize the extraction process of polysaccharides from Pholiota namek by ultrasonic synergistic.The DPPH·, O2~-·and·OH scavenging ability and reducing power of polysaccharides from Pholiota namek were studied.The polysaccharide yield was determined by thephenol-sulphuric acid method with hot water extraction after ultrasonic.Through the single factor test and orthogonal, the effect of polysaccharide yield by ultrasonic power, ultrasonic time and ratio of water to raw material were researched.The best process was 80Hz, 15min and 20∶ 1, respectively.The results indicated that Pholiota nameko polysaccharide had better antioxidant activity via evaluating free radicalscavenging and reducing power.
