牛蒡提取物对刀豆蛋白A诱导肝损伤小鼠的保护作用

Hepatoprotective effect of burdock concentrate against ConA induced liver damage in mice

  • 摘要: 目的:研究牛蒡提取物对刀豆蛋白A(ConA)诱导肝损伤小鼠的保护作用并探讨其可能的作用机制。方法:小鼠尾静脉注射ConA建立肝损伤模型。计算小鼠肝脏、脾脏和胸腺指数,测定血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)和肝组织中肿瘤坏死因子(TNF-α)和一氧化氮(NO)的含量。观察肝组织病理学变化。结果:牛蒡皮乙醇提取物(PA)显著改善肝损伤小鼠的肝脏和胸腺指数(p<0.05);降低血清中ALT、AST活性和肝组织匀浆中TNF-α、NO的含量(p<0.01);肝脏病变明显减轻。去皮牛蒡根水提物(MA)未表现出护肝作用。结论:牛蒡皮乙醇提取物(PA)对ConA诱导肝损伤小鼠具有保护作用,其作用机制可能与抑制T淋巴细胞的活化和相关炎症因子的释放相关。 

     

    Abstract: Purpose:To investigate the protective effects of burdock concentrate on concanavalin A (ConA) -induced liver injury in mice.Methods:Mice liver injury model was induced by ConA vena caudalis injection.Liver, thymus and spleen index, contents of ALT, AST in serum and TNF-α, NO in liver were measured.Liver histopathological changes were examined accordingly.Results:Compared with modal group, the liver index was decreased and thymus index was increased in PA (peel of burdock extrated by ethanol) group (p<0.05) .The contents of ALT, AST in serum, and TNF-α, NO in liver homogenate were significantly decreased (p<0.01) .A significant improvement was also observed in pathohistology examination.MW (peeled root extrated by water) had no effect on biochemical indicators and liver histological morphology in mice.Conclusion:PA has protective effect on ConA-induced liver injury in mice.This maybe implementated by inhibiting T lymphocyte activity and decreasing the release of inflammatory factors.
