稠李属三种果实花色苷对HepG2细胞的增殖抑制作用比较

Effect of anthocyanins extracted from three kinds of Padus Mill fruits on the proliferation of HepG2 cell

  • 摘要: 目的:比较稠李属三种果实花色苷对人肝癌HepG2细胞增殖抑制作用。方法:质量浓度为0.05~1.2mg/mL的稠李属三种果实花色苷分别作用于HepG2细胞24、48、72h,采用胎盘蓝染色法和MTT法绘制生长曲线以及检测对HepG2细胞生长抑制率。结果:0.05mg/mL的紫叶稠李花色苷促进细胞增殖,而山桃稠李、稠李在试选浓度范围内均对HepG2细胞具有抑制作用,且随浓度增加及作用时间延长抑制率增大。在紫叶稠李、山桃稠李、稠李作用HepG2细胞48h时,IC50值依次为1.07、0.82、0.64mg/mL,显示抑制HepG2细胞增殖的大小关系为:稠李>山桃稠李>紫叶稠李。结论:稠李属的果实花色苷能够有效抑制肝癌HepG2细胞的增殖。 

     

    Abstract: Objective:Research the inhibition of anthocyanins extracted from three kinds of Padus Mill fruits on the proliferation of HepG2 cell.Methods:The anthocyanins of different mass concentration extracted from three kinds of Padus effect on HepG2 cell for 24, 48 and 72h, then by placental blue staining method to calculate the number of active cells, and draw the proliferation curves.Test the inhibition rate of anthocyanins on the proliferation of cell by the method of MTT.Result:The anthocyanins of Padus virginiana increased the cell proliferation at the mass concentration of 0.05mg/mL.The anthocyanins of Padus racemosa and Prunus maackii caused the inhibition of cell proliferation in the selected concentration range, and the inhibition increased as the mass and time increased.After 48 hours, the number of IC50of the HepG2 cell to the anthocyanins was Padus Virginiana:1.07mg/mL, Prunus maackii:0.82mg/mL, Padus racemosa:0.64mg/mL, the order of inhibition was Padus racemosa>Prunus maackii>Padus Virginiana.Conclusion:The anthocyanins of three kinds of Padus Mill fruits caused inhibition on the proliferation of HepG2 cell effectively.
