假单胞菌BL11产碱性脂肪酶发酵条件的优化

Fermentation optimization of alkaline lipase produced by the pseudomonas BL11

  • 摘要: 通过单因素实验和均匀设计对已分离到的一株假单胞菌BL11产碱性脂肪酶的培养基配方和摇瓶发酵条件进行初步优化。优化后的最佳培养基组成为(w/v):麦芽糖2.5%,蛋白胨3.0%,豆油0.5%,K2HPO40.2%;最佳发酵条件为:起始pH7.5,装液量40mL/250mL三角瓶,培养温度33℃,摇床转速180r/min,发酵周期60h。在上述条件下,BL11菌株产脂肪酶酶活可高达223U/mL。 

     

    Abstract: The optimal culture medium and culture conditions of alkiline lipase produced by the pseudomonas BL11 was studied by single-factor experiment and uniform design.The optimized medium was maltose 2.5%, peptone 3.0%, soybean oil 0.5%, K2HPO4 0.2% and the optimized culture conditions was initial pH7.5, medium volume 40mL/250mL flask, culture temperature 33℃, rotatiaon rate 180r/min.The highest enzyme activity was 223U/mL after 60h.
