表面活性剂与β-环化糊精共同作用对放线菌羟基化维生素D3的影响

Effect of surfactant and β-cyclodextrin on vitamin D3 Hydroxylased by Actinomyces

  • 摘要: 放线菌CXX-10具有维生素D3的25位羟基化活性。通过研究Tween-80、聚乙二醇辛基苯基醚(Triton-100)、甘油、十二烷基硫酸钠(SDS)、十六烷基溴化铵(CTAB)等表面活性剂,与β-环化糊精共同作用对该菌25位羟基化维生素D3的影响,获得了较优的促发酵组合并优化了其反应条件:培养3d后添加0.04% Tween-80、0.5% β-环化糊精和VD3,pH为7.0,28℃,180r/min,发酵转化反应7d,25-(OH)D3产率达14.5mg/L。 

     

    Abstract: Actinomyces CXX-10 is capable of 25-hydroxylating of vitamin D 3 . The effect of surfactant such as Tween-80, Triton-100, glycerin, sodium dodecyl sulfate (SDS) , hexadecyl trimethyl ammonium Bromide (CTAB) cooperate with β-cyclodextrin on productivity of 25 - (OH) D 3 was analysised in this paper. And then optimization of condition of most suitable combination of surfactant and β-cyclodextrin were promoted. The optimal conditions were as follows:0.04% Tween-80 and 0.5% β-cyclodextrin and V D 3 were added after 3d of culture, and other culture condition were 28℃, 180r/min, 7d.
