响应面法优化超声波辅助提取葡萄悬浮细胞花色苷

Optimization of ultrasound-assisted extraction of anthocyanin in Vitis vinifera suspension cultures by response surface methodology

  • 摘要: 采用响应面法对葡萄悬浮细胞花色苷的提取工艺参数进行优化。在单因素实验基础上,选取酸化乙醇浓度(含体积分数为0.1%的盐酸)、超声功率、提取时间和提取温度为自变量,以花色苷含量为响应值,利用Box-Behnken中心组合设计原理和响应面分析法,研究各自变量及其交互作用对花色苷含量的影响,并优化提取工艺。结果表明,提取的最佳工艺条件为:料液比1∶9、酸化乙醇浓度85%、超声功率300W、提取时间38min、提取温度42℃。在此条件下获得的花色苷提取产量预测值为3.96mg/L,与实测值3.93mg/L相符。超声波技术适用于葡萄悬浮细胞花色苷的提取,所拟合的统计模型可信度高。本研究结果对工厂化生产葡萄花色苷的研究具有较好的借鉴意义。 

     

    Abstract: Response surface methodology ( RSM) was used to optimize extraction of anthocyanin from Vitis vinifera suspension cultures.Based on single factor tests, acidic ethanol concentration ( containing 0.1% hydrochloric acid, V/V) , ultrasonic power, extraction time and extraction temperature were chosen as arguments and the content of anthocyanin as response value.The interaction of the respective variables and their influence on the content of anthocyanin and the optimum process were studied by using Box- Behnken central composite design and response surface analysis theory.The results showed that the optimum conditions were: liquid- solid ratio of 1∶9, acidic ethanol concentration 85%, ultrasonic power 300W, extraction time 38min and extraction temperature 42℃ for twice.Under these conditions, the content of anthocyanins was up to 3.96mg /L, which was consistent with the measured value ( 3.93mg /L) .The results indicated that ultrasound technology was suitable for the extraction of anthocyanins in Vitis Vinifera suspension cultures, and the statistical model fitting was with high reliability.This study has some practical significance for the research of the industrialized production of anthocyanins.
