不同食用油脂对大鼠血脂代谢及脂肪肝的影响

Effect of different oil on lipid metabolism and hepatic steatosis in rats

  • 摘要: 研究不同油脂(脂肪酸构成比例)对大鼠血脂及脂肪肝的影响。将SPF级SD雄性大鼠分别分为五组,每组7只,正常对照组喂以基础饲料,高脂组喂以含不同油脂(猪油、棕榈油、橄榄油、油茶籽油)的高脂饲料,测量体重、肥胖指数,大鼠取血处死后,分离血清检测血脂和肝功能,摘取肝脏称重并留取肝脏组织固定做HE染色。结果显示,高脂饲料喂养17周后,各油脂组大鼠出现不同程度的肥胖,表现为体重、体脂、Lee’s指数明显增加;猪油组肝脏指数略高于其他组,但病理切片显示猪油组脂肪肝现象最为明显,橄榄油和油茶籽油组亦有轻度脂肪肝表现。血脂分析显示猪油组血脂最高,其次为棕榈油,橄榄油和茶籽油组血脂升高不显著,茶籽油组血脂与正常组无差异;猪油组氧化应激程度最明显,其次为棕榈油,橄榄油和油茶籽油组与正常对照组差异不大。提示SFA能升高血脂,进而引起氧化应激,导致肝损伤;而UFA与适当n-6/n-3PUFA比值够调节血脂代谢和减少氧化作用,从而减轻脂肪肝程度。 

     

    Abstract: To investigate effect of different oil ( different proportion of fatty acid composition) on lipid metabolism and hepatic steatosis in rats.Male SD rats in a SPF grade were randomly divided into five groups ( N = 7) .Rats in control group were fed a standard chow diet and rats in model groups were fed high fat diet containing different oil ( lard, palm oil, olive and camellia seeds oil) .In the end of 17th week, body weight and obesity index were measured.After the rats were sacrificed, serum lipids and liver function were analyzed.The livers were weighed and fixed in formalin to make HE stained biopsy.The results showed that chronic high fat diet induced obesity, with significantly increased body weight, fat and Lee's index.The lard group exhibited a slightly higher liver index and the most serious hepatic steatosis compared with the other groups.While the olive oil and camellia seed oil groups revealed light degree of hepatic steatosis.Among the four high fat diet groups, the rats in lard fed group revealed marked hyperlipemia, followed by the palm oil group.There was no significant change in the serum lipids of the camellia seed oil and olive oil diet groups and it showed no difference between camellia seed oil and the control group.The serum SOD, MDA and LPO exhibited the same trend as serum lipids in the four high fat diet groups.The above results suggest that SFA can increase hyperlipemia, thereby causing oxidative stress and leading to liver damage.The amount of UFA along with a proper n-6 /n-3PUFA ratio can regulate lipid metabolism and decrease oxidative stress, consequently reducing hepatic steatosis.
