不同方法提取国槐叶黄酮及其抗氧化活性研究

Study on the extraction method and antioxidant activity of flavonoids from Sophora japonica L.

  • 摘要: 研究超声波辅助双水相、回流、半仿生、水提、超声波辅助双水相(碱)性5种提取方法提取国槐叶黄酮以及抗氧化活性。结果表明,回流提取黄酮含量大于其它几种提取方法,与其它3种方法比较黄酮含量差异显著(p<0.05),而与超声波辅助双水相提取比较差异不显著(p>0.05)。抗氧化结果显示,5种提取方法所得的黄酮提取物均有较强的抗氧化活性,但超声波辅助双水相黄酮提取物对DPPH自由基的清除作用和对铜离子的还原力优于其它方法,超声波辅助双水相(碱)性黄酮提取物对ABTS自由基的清除作用及对铁离子的螯合能力最好。而不同提取物对亚油酸自氧化的抑制作用的强弱顺序是回流>超声波辅助双水相>半仿生>超声辅助双水相(碱)>水提。由此说明,国槐叶黄酮易溶于有机溶剂,超声辅助双水相提取物具有很好的抗氧化性。 

     

    Abstract: The study aimed to explore the flavonoids contents and antioxidant activity of diffrerent extracts from Sophora japonica L by ultrsonoc-assisted aqueous two-phase extraction ( UATPE) , reflux extraction ( RE) , Semibionic extraction ( SBE) , water extraction ( WE) , Ultrasonic assisted aqueous two- phase ( alkaline UATPEA) , respectively.The results indicated that the content of flavonoids extracted by RE was the highest, which showed a significant difference ( p < 0.05) compared with other methods except for UATPE ( p > 0.05) .The antioxidant results showed that flavonoids extracts by the five different methods obtaining had strong antioxidant activity.The scavenging ability of DPPH free radical and reduction of copper ions of flavonoids extracted by UATPE was the strongest.In addition, ABTS radical scavenging effect and iron ion chelating power of UATPEA flavonoids extractes were better than that of other methods.Moreover, the extracts obtained by different methods inhibition linoleic acid was RE > UATPE > SBE > UATPEA > WE. The results indicated that the flavonoids in Sophora japonica L were soluble in organic solvent.The extracts obtained by UATPE had better antioxidant activity.
