丙酮磷酸氢二钾双水相体系提纯黄褐毛忍冬中的绿原酸

Separation and purification of chlorogenie acid from Lonicera fulvotomentosa with aqueous two-phase systems containing acetone and Dipotassium Hydrogen Phsophate

  • 摘要: 本研究采用丙酮-磷酸氢二钾双水相体系法提纯黄褐毛忍冬中的绿原酸,通过正交实验测定了酮水比、盐液比和料液比对绿原酸萃取率及提取物纯度的影响。结果表明:按酮水比8.5∶1.5(V/V),盐液比为100∶1(mg/mL)配制双水相,料液比25∶1(mg/mL),绿原酸的萃取率为91.10%,纯度从原料中的20.16%提高到42.13%。该工艺溶剂易回收、易处理、操作简便,适于黄褐毛忍冬中绿原酸的提纯。 

     

    Abstract: Chlorogenie acid was separated and purified from Lonicera fulvotomentosa crude extract by acetoneK2HPO4two-phase aqueous system.Influence of material /liquid ratio, acetone /water ratio and salt /liquid ratio on extraction rate and purity of chlorogenie acid was investigated by orthogonal test. The results showed that extraction yield of chlorogenie acid was 91.10% under acetone- water ratio for 8.5∶1.5 ( V/V) , salt- solvent ratio for100∶1 ( mg /mL) and material-solvent ratio for 25∶1 ( mg /mL) , the purity of chlorogenie acid increases from 20.16%with crude extract 42.13%. This method was easy to recovered solvent, dispose and operate, which fit for purification of chlorogenie acid from Lonicera fulvotomentosa.
