常压室温等离子体快速诱变选育丙酮酸高产菌株

Rapid breeding of a high pyruvate production mutant by atmospheric room temperature plasma

  • 摘要: 本研究以产丙酮酸的光滑球拟酵母(Torulopsis glabrata)Tp19为出发菌株,采用常压室温等离子诱变系统(ARTP)进行诱变,根据CaCO3筛选平板透明圈与菌落直径比以及100孔板产酸量快速筛选丙酮酸高产突变株。最后通过摇瓶发酵复筛,选育出一株遗传稳定的突变株A214,丙酮酸产量达到37.43g/L,比出发菌株提高了13.69%。 

     

    Abstract: A novel mutation approach, the atmospheric room temperature plasma (ARTP) , was used to treat Torulopsis glabrata Tp19 for the breeding of high pyruvate producer. The mutant with high pyruvate production was quickly screened out based on the ratio of transparent circle to colony diameter on CaCO3 screening plate with colony diameter and the acid production on 100-well plate. The obtained mutant A214 exhibits good genetic stability and pyruvate production reached 37.43g/L, which was 13.69% higher than that of original strain.
