新疆新春36号黑小麦中硒的分布及富硒蛋白提取工艺研究

Study on the distribution and the extraction process of rich proteins of Xinchun No.36 Triticale in Xinjiang province

  • 摘要: 为了探究新疆新春36号黑小麦中硒的分布,本实验通过单因素及响应面分析法对硒蛋白、硒多糖、硒核酸及不同溶解性的硒蛋白做了系统考察。结果表明:36号黑小麦中硒蛋白、硒多糖及硒核酸中硒的含量分别为(240.52±0.3)、(32.45±0.21)、(4.65±0.05)mg/g;碱溶性、水溶性、醇溶性和盐溶性硒蛋白的含量分别为(146.21±0.3)、(41.52±0.6)、(19.40±0.06)、(13.65±0.12)mg/g。碱溶性硒蛋白的最佳提取条件为:Na OH溶液浓度0.17 mol/L,酸化p H4.7,(NH4)2SO4饱和度为66.0%,料液比为1∶10。该条件下模型预测碱溶性硒蛋白的提取率为76.5593%,实际实验值为76.7%。 

     

    Abstract: The purpose of this work was to study the distribution of Se in Xinchun No.36 triticale in Xingjiang province. Through single factor experiment and response surface method, a systematic inspection of selenoproteins,selenium polysaccharide,selenium nucleic acid and selenoproteins of solubility in different solvents was made. The results showed that content of selenoproteins,selenium ploysaccharide and nucleic acid selenium was(240.52±0.3),(32.45±0.21) and(4.65±0.05) mg/g respectively. The content of selenoproteins of alkali-soluble,water-soluble,salt-soluble and alcohol-soluble was(146.21±0.3),(41.52±0.6),(19.40±0.06)and(13.65±0.12) mg/g respectively. Furthermore,in order to get more alkali-soluble selenoproteins,the optimal extraction technology was obtained by the single factor test and the response surface design experiments. The optimal extraction conditions were Na OH 0.17 mol/L,acidification at p H4.7,saturation of(NH4)2SO466.0%,the ratio of solid to liquid 1 ∶10, the predicted rate of protein extraction was 76.5593%, but the actual value of experiment reached 76.7%.
