  • 摘要: 乳糜泻关联表位的定量检测可以评价小麦种质资源及加工食品的乳糜泻致敏性,为患者安全消费提供技术保障。定量检测小麦麸质蛋白中致乳糜泻表位的方法包括免疫印迹法、高通量测序法和多反应检测串联质谱检测(LC-MRM/MS),它们在麸质表位的检测中各具特色。免疫印迹法检测结果稳定直观,但定量精确度不高;高通量测序法准确度相对较高,但应用范围窄,且数据处理困难;多反应检测串联质谱检测灵敏度高、样品用量少、分析速度快,但费用较高。 

     

    Abstract: Detection of epitopes in gluten related to celiac disease can evaluate the sensitivity of wheat variety resources and the relevant food,and it is a strong technical support for the safety consumption for celiac disease patients.The main methods used in the detection of epitopes related to celiac disease in wheat gluten involve three approaches based on western blotting,next- generation sequencing,LC- MRM / MS,respectively.The advantages and disadvantages of these methods were introduced in detail.Among them,western blotting could present direct results,while the quantitative accuracy was not high.The accuracy of next- generation sequencing is relatively high,while the range of application is narrow with a complicated data processing.LC- MRM / MS has advantages on high sensitivity,low sample consumption and fast analysis speed with higher cost.
