高效液相色谱法测定葡萄皮、籽中的15种单体酚

Determination of 15 mono- phenols in grape skin and seeds by high performance liquid chromatography

  • 摘要: 建立了葡萄皮、籽中15种单体酚的高效液相色谱(HPLC)分析方法,采用ZORBAX SB-C18(4.6 mm×250 mm,5μm)色谱柱,柱温30℃,流动相乙腈-甲酸水(2∶98 V/V)溶液,梯度洗脱,洗脱流速为1.0 m L/min,检测波长280 nm。15种单体酚在一定浓度范围内呈良好的线性关系,相关系数为0.99800.9999。方法检出限0.010.10 mg/L,加标回收率均大于85%。本方法快速、准确、灵敏度高,可用于葡萄皮、葡萄籽中15种单体酚的测定。 

     

    Abstract: A high performance liquid chromatograohic( HPLC) method was established for the determination of 15mono-phenols in grape skin and grape seeds. The ZORBAX SB-C18( 4.6 mm × 250 mm,5 μm) was used as chromatographic column with column temperature of 30 ℃.Acetonitrile and formic acid aqueous solution was used as the mobile phase by gradient elution with a flow rate of 1.0 m L/min. The detection wavelength was set at280 nm. A good linearity was obtained for 15 mono-phenols in the range of 0.01 ~ 0.10 mg/L. The correlation coefficients were in range of 0.9980 ~ 0.9999. The recoveries of standard samples were in the range of 93.25% ~109.12%. The method was rapid,simple and high senstivity and was suitable for the determination of 15 monophenols in grape skin and grape seeds.
