青龙衣提取物的抗真菌和抗肿瘤活性研究

Antifungal and antitumor activity of walnut green husk'extracts

  • 摘要: 将青龙衣乙醇提取物用石油醚、氯仿、乙酸乙酯、正丁醇四种不同极性的有机溶剂依次萃取,以4种玉米主要病原真菌和2种肿瘤细胞对青龙衣提取物进行抗真菌和抗肿瘤活性研究。结果表明:乙醇提取物及其不同极性萃取相对供试病原真菌和肿瘤细胞均有一定的抑制作用。其中青龙衣乙醇提取物对玉米穗腐菌、玉米黑粉菌、玉米弯孢菌和玉米纹枯菌等真菌的抑菌率分别为92.5%、86.0%、83.2%、50.0%,乙酸乙酯萃取相的抑菌效果最好,其EC50分别为7.354、9.546、10.341和96.532 mg/m L;MTT比色法实验表明:不同萃取相对Hela细胞和Huh7.5.1细胞的抑制呈现剂量-效应关系,其中石油醚萃取相对两种细胞的抑制效果最明显,其对Hela和Huh7.5.1细胞的IC50分别为124.99μg/m L和45.33μg/m L,并可诱导Huh7.5.1细胞凋亡。说明青龙衣提取物对玉米穗腐菌、玉米黑粉菌、玉米弯孢菌、玉米纹枯菌和Hela细胞、Huh7.5.1细胞均有抑制作用,具有剂量依赖性。 

     

    Abstract: The ethanol extracts of walnut green husk were further extracted with petroleum ether,chloroform,ethyl acetate and n- butanol.Antifungal and antitumor activity of the walnut green husk'different solvent extracts against4 corn pathogenic fungi and 2 tumor cells were studied. The results showed that the ethanol extract from walnut green husk and different solvent extracts had certain inhibition on testing microbial.For the corn pathogenic fungi,the inhibition rate of ethanol extract to Fusarium graminearum,Curvularia lunata,Rhizotonia solani Kuhn and Ustilago maydis were 92.5%,86.0%,83.2%,50.0% respectively.The ethyl acetate extract had the best antimicrobiol activity among the different solvent extracts,which the EC50 were 7.354,9.546,10.341,96.532 mg / m L. MTT assay showed that the different extracts inhibited the growth of Hela cells and Huh7.5.1 cells in a dose- dependent manner.The petroleum ether extract had the most obvious antitumor effect on testing cells,which the IC50 were124.99 μg / m L and 45.33 μg / m L,it could induce Huh7.5.1 cell's apoptosis. The ethanol extracts of walnut green husk can inhibit Fusarium graminearum,Curvularia lunata,Rhizotonia solani Kuhn,Ustilago maydis and Hela cells,Huh7.5.1 cells in a dose- dependent manner.
