Bohai sea-9145重组脂肪酶基因工程菌的发酵表达条件优化

Optimization of fermentation and expression conditions for production of marine lipase by Bohai sea-9145

  • 摘要: 为提高大肠杆菌(E.coli)基因工程菌产Bohai sea-9145重组脂肪酶(Lipase,EC3.1.1.3)的能力,有效制备Bohai sea-9145脂肪酶。采用单因素实验、Plackett-Burman实验和Box-Behnken响应面设计实验,对该重组酶基因工程菌的发酵表达条件进行了优化。最终确定的最佳条件是:接种量为1.5%,培养基的初始p H为8.0,诱导剂IPTG的添加时间为3.5 h,终浓度为0.5 mmol/L,诱导时间为15 h,诱导温度为16℃,摇床转速为200 r/min,经优化后该菌的产酶量是优化前的9.72倍。 

     

    Abstract: To effectively improve the production of the Bohai sea- 9145 lipase,single factor experiment,PB experiment and BB surface response experiment were used to optimize the fermentation conditions of the recombinant strain Bohai sea-9145.The optimal conditions were as follows: the initial p H value of the medium was8.0,the inoculation amount was 1.5%,and after 3.5 hours of incubation,the expression of the lipase was induced by adding IPTG to a final concentration of 0.5 mmol / L at 16 ℃ for 15 hours with the shaking speed of 200 r / min.After optimization,the final activity of the recombinant lipase was increased by 9.72 times.
