高效液相色谱-蒸发光散射法测定食品中糖醇含量的研究

Study on the determination of sugar alcohols in foods by high performance liquid chromatography-evaporative light scattering detection

  • 摘要: 针对口香糖、饼干、面包、饮料等食品中添加的代糖成分赤藓糖醇、木糖醇、山梨糖醇和麦芽糖醇的检测,建立了水和乙腈为流动相,等度洗脱,氨基色谱柱分离,蒸发光散射检测器检测食品中添加的糖醇含量的高效液相色谱方法。赤藓糖醇、木糖醇、山梨醇和麦芽糖醇的线性工作范围分别为140~525、100~500、100~500、100~500μg/m L,线性相关系数R2分别为0.998、0.999、0.999和0.999,对不同类型的食品,四种糖醇的回收率在91.9%~105.2%之间,方法准确度高。室内重复性变异系数一般都低于5%,室间再现性变异系数一般都低于10%,说明方法的重复性和再现性良好。该方法简单、便于操作,测定结果准确,适于复杂食品基质中糖醇含量的测定。 

     

    Abstract: A high performance liquid chromatography- evaporative light scattering detection( HPLC- ELSD) method had been developed for the determination of erythritol,xylitol,sorbitol and maltitol in chewing gum,biscuits,bread and beverage.The sugar alcohols in foods were separated on amino column by isocratic elution with acetonitrilewater solution( 80∶ 20,V∶ V). The linear range of standard working curve for erythritol,xylitol,sorbitol and maltitol were 140~525 μg / m L,100~500 μg / m L,100~500 μg / m L and 100~500 μg / m L,with linear correlation coefficient( R2)of 0.998,0.999,0.999 and 0.999,respectively.For different kinds of food,the recovery of four kinds of sugar alcohols were in the range of 91.9% ~ 105.2%. The coefficient of variations of intra- laboratory repeatability and inter-laboratory reproducibility were less than 5% and 10%,respectively. This method was simple,easy to grasp and accurate in the determination of sugar alcohols in complex food matrix.
