响应面法优化红芸豆种皮花色苷的超声波辅助提取工艺及其抗氧化活性研究

Optimization of ultrasonic assisted extraction of anthocyanins from seed coat of red kidney bean ( Phaseolus vulgaris L.) using response surface methodology and evaluation of its antioxidant activity

  • 摘要: 为了探索新的花色苷资源,提高红芸豆的价值。本文在单因素实验的基础上应用响应面法对红芸豆种皮花色苷的超声波辅助提取条件进行优化并评估其体外抗氧化活性。最佳条件如下:采用40%乙醇溶液(HCl调p H为1),料液比1∶24(g/m L),提取温度为68℃,采用160 W超声处理78 min时花色苷得率最高达到185.57 mg/100 g;抗氧化活性实验表明红芸豆种皮花色苷提取物对ABTS+·、DPPH·的最高清除率分别达到68.11%、90.66%,同时具有较高的总还原能力。优化的红芸豆种皮花色苷提取工艺科学、合理,红芸豆种皮花色苷提取物具有较强的抗氧化活性。 

     

    Abstract: In order to explore new anthocyanins resources, the ultrasonic assisted extraction of anthocyanins from seed coat of red kidney bean ( Phaseolus vulgaris L.) was optimized by response surface methodology based on the single factor experiments and the antioxidative activity of the crude extract was evaluated. According to the results, the optimal extraction solvent was40% ethonal ( modified to p H1 by hydrochloric acid) , and the highest extraction yield of 185.57 mg anthocyanins/100 g seed coat powder was achieved at an extraction temperature of 68 ℃, an ratio of raw material to solvent of 1 ∶ 24 ( g/m L) , an extraction time of 78 min and the ultrasonic power was 160 W.The results of antioxidation experiments showed that the highest scavenging capacity of anthocyanins against ABTS free radical, DPPH free radical was 68.11%, 90.66% respectively and the ferric reducing antioxidant power was in direct proportion to the anthocyanins concentration. The optimization of ultrasonic extraction of anthocyanins from seed coat of red kidney bean was reasonable and scientific, and the anthocyanins had stronger antioxidant activity.
