水提铵沉法提取蜂王浆水溶性蛋白工艺优化

Optimization of water-soluble protein extraction process of royal jelly by water extraction and ammonium precipitation

  • 摘要: 选定黑龙江密山蜂王浆作为原料,以水溶性蛋白提取率作为评价指标。在单因素实验和PB实验的基础上,利用响应面优化法对水提铵沉法提取蜂王浆水溶性蛋白的工艺进行优化。实验结果显示,最佳工艺参数为料水质量比1∶20、超声波提取时间40 min、硫酸铵饱和度89%、pH5、沉淀时间50 min,此条件下蜂王浆水溶性蛋白提取率达到80.22%,是适用于蜂王浆水溶性蛋白提取的较好方法。 

     

    Abstract: This experiment chose the royal jelly from Heilongjiang Mishan as raw material and took water-soluble protein extraction rate as evaluation indicator.Based on single factor experiment and Plackett-Burman design, the response surface optimization method was used to optimize the extraction of water-soluble protein from royal jelly by water extraction ammonium sulfate precipitation.The experiment showed that the optimum technological parameters were material water mass ratio 1 ∶ 20, ultrasonic extraction time 40 minutes, ammonium sulfate saturation 89%, pH5, precipitation time 50 minutes.Under the condition of this, the water-soluble protein extraction rate of royal jelly was up to 80.22%, this method of experiment was the better way to extract water-soluble protein of royal jelly.
