高效液相色谱法同时测定酵素原液中的乳酸和醋酸

Simultaneous determination of lactic acid and acetic acid in enzyme solution by high performance liquid chromatography

  • 摘要: 建立了一种利用高效液相色谱法同时测定酵素原液中乳酸和醋酸的方法。通过对检测波长、流动相及其浓度和pH的筛选,建立利用高效液相色谱法同时测定酵素原液中乳酸与醋酸含量方法。结果表明,最优色谱条件为:色谱柱SinoChrom ODS-BP(4.6 mm×150 mm,5 μm),流动相为0.01 mol/L、pH2.88的KH2PO4溶液,流速0.8 mL/min,波长215 nm,进样量20 μL,柱温25 ℃。该法可同时测定酵素中乳酸和醋酸,检测速度快,乳酸与醋酸的线性检测范围宽分别为0~2100、0~1800 μg/mL。精密度和重复性好(乳酸和醋酸平行测定的RSD分别为0.99%、1.21%)并得到了良好的回收率(92.31%~102.20%)。

     

    Abstract: A method for simultaneous determination of lactic acid and acetic acid in jiaosu solution by high performance liquid chromatography (HPLC) had been established. This article focused on the establishment of the simultaneous determination method of lactic acid and acetic acid in jiaosu solution by high performance liquid chromatography through the optimization of the detection wavelength, mobile phase and its concentration and pH. The results showed that the optimal Chromatographic conditions are as follows:Chromatographic column (SinoChrom ODS-BP 4.6 mm×150 mm, 5 μm), 0.01 mol/L KH2PO4 solution (pH2.88) as mobile phase, flow rate of 0.8 mL/min, wavelength of 215 nm, injection volume of 20 μL, column temperature of 25℃. The method could be used for simultaneous determination of lactic acid and acetic acid in jiaosu solutions. The method has the advantages of fast detection, wide linear detection range of lactic acid (0~2100 μg/mL) and acetic acid (0~1800 μg/mL), good precision and repeatability (the RSD of parallel determination of lactic acid and acetic acid were 0.99% and 1.21%, respectively) and good recoveries (92.31%~102.20%).
