基于气相色谱-质谱联用技术的平菇多糖治疗大鼠急性肝损伤的代谢组学研究

Metabolomics Study of Pleurotus ostreatus Polysaccharides on the Treatment of Acute Liver Injury in Rats based on Gas Chromatography-Mass Spectrometry

  • 摘要: 目的:研究平菇多糖对急性肝损伤大鼠尿液代谢组学的影响。方法:以四氯化碳造大鼠急性肝损伤动物模型,采用生化分析仪测定血中谷丙转氨酶(ALT)、谷草转氨酶(AST)和碱性磷酸酶(ALP)水平,采用气相色谱-质谱联用技术研究平菇多糖对肝损伤大鼠尿液代谢标志物和代谢通路的影响。结果:与模型对照组比较,平菇多糖组能显著降低血中ALT、AST和ALP水平(p < 0.05);显著升高尿液代谢物柠檬酸、苹果酸、D-葡萄糖醛酸和1,2,3-丙烷酸水平(p < 0.05),显著降低十六烷酸水平(p < 0.05);这些标志物属于三羧酸循环、乙醛酸和二羧酸代谢、抗坏血酸和醛酸代谢。结论:平菇多糖能改善肝损伤大鼠的症状,调节肝损伤病大鼠尿液柠檬酸、苹果酸、D-葡萄糖醛酸、十六烷酸和1,2,3-丙烷酸水平以及三羧酸循环、乙醛酸和二羧酸代谢、抗坏血酸和醛酸代谢通路异常,具有明显的保肝作用,将其开发为保肝功能性食品有一定的应用价值。

     

    Abstract: Objective:This study was to evaluate the hepatoprotective effect of polysaccharides from Pleurotus ostreatus (POP) on acute liver injury rats.Methods:An animal model of acute liver injury in rats was induced by carbon tetrachloride.The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) in blood were determined by biochemical analyzer.The effects of Pleurotus ostreatus polysaccharides on urinary metabolic markers and metabolic pathways in rats with liver injury were analyzed by gas chromatography-mass spectrometry.Results:The levels of ALT, AST and ALP were markedly decreased in POP group compared with model group (p < 0.05).Meanwhile, after treatment with POP, the levels of citric acid, malic acid, D-glucuronic acid and 1, 2, 3-propanetricarboxylic acid decreased (p < 0.05) and hexadecanoic acid level increased (p < 0.05). These biomarkers belonged to TCA cycle, glyoxylate and dicarboxylate metabolism, ascorbate and aldarate metabolism. Conclusion:POP could ameliorate the abnormal changes of liver function and regulate the disorder of biomarkers and pathways on acute liver injury rats.Conclusion:POP had a hepatoprotective effect on acute liver injury and had certain application value to be developed as liver protection functional foods.
