两株植物乳杆菌联合抑制高脂诱导小鼠肥胖的形成

Two Lactobacillus plantarum Combined to Inhibit the Formation of Obesity Induced by High Fat in Mice

  • 摘要: 目的:考察植物乳杆菌KLDS1.0344和KLDS1.0386联合处理对小鼠肥胖形成的抑制作用。方法:实验小鼠分为空白组、模型组、干预组和药物组,空白组喂养D12450B对照饲料,其余三组喂养D12492高脂饲料,空白组和模型组灌胃灭菌PBS缓冲溶液,干预组(KLDS1.0344组、KLDS1.0386组、混合组)和药物组分别灌胃菌悬液0.2 mL(混合组按照1:1比例,菌浓度为5×108 CFU/mL)、奥利司他(10 mg/kg),持续8周,测定小鼠体重、Lees系数、体脂率、血脂四项、肝脏甘油三酯、肝脏HE切片等相关生化指标。结果:8周后,模型组体重高于空白组27.5%,模型组建成功;与模型组体重增量10.49 g相比,混合组体重增量5.36 g显著下降(P<0.05),Lees系数3.12%、体脂率2.33%、血脂(TC:4.23 mmol/L、TG:0.80 mmol/L)及肝脏甘油三酯含量(0.28 mmol/g prot)显著下降(P<0.05);从肝脏切片中可以观察到模型组小鼠肝脏中出现明显的脂肪变性,而干预组中显著缓解(P<0.05)。结论:该联合菌株可作为抑制高脂诱导肥胖形成的潜在益生菌制剂。

     

    Abstract: Objective:To investigate the inhibitory effect of Lactobacillus plantarum KLDS1.0344 and KLDS1.0386 on obesity formation in mice. Methods:The experimental mice were divided into blank group,model group,intervention group and drug group. The blank group was fed D12450B control feed,the other three groups were fed D12492 high fat diet,the blank group and the model group were intragastrically sterilized with PBS buffer solution and intervention group(KLDS1.0344 group,KLDS1.0386 group,mixed group)and drug group were fed 0.2 mL gastric suspension(mixed group according to 1:1 ratio,bacterial concentration 5×108 CFU/mL),orlistat(10 mg/kg),respectively for 8 weeks,the body weight,Lees coefficient,body fat percentage,blood lipids,liver triglycerides,liver HE slices and other related biochemical indicators were determined. Results:After 8 weeks,the weight of the model group was higher than that of the blank group by 27%,and the model was successfully established. Compared with the weight gain of the model group of 10.49 g,the weight gain of the intervention group was significantly decreased by 5.36 g(P<0.05),and the Lees coefficient was 3.12%. Body fat percentage was 2.33%,blood lipids(TC:4.23 mmol/L,TG:0.80 mmol/L)and liver triglyceride content(0.28 mmol/g prot)were significantly decreased(P<0.05);models were observed from liver sections. Significant steatosis occurred in the liver of the group,and significant remission in the intervention group(P<0.05). Conclusion:This combined strain can be used as a potential probiotic preparation to inhibit the formation of high fat-induced obesity.
