长白落叶松3种黄酮化合物定性、定量分析及其抗氧化活性研究

Qualitative and Quantitative Analysis of 3 Kinds of Flavonoids in Larix olgensis var. koreana Extract and Their Antioxidant Activities

  • 摘要: 以我国东北地区长白落叶松锯末子为研究对象,通过HPLC-MS/MS方法定性定量分析长白落叶松中主要黄酮成分及含量,并测定其抗氧化活性。结果显示,长白落叶松主要含三种黄酮,分别为二氢槲皮素(92.07%)、香橙素(2.39%)和圣草酚(0.19%)。抗氧化活性实验结果表明,长白落叶松黄酮提取物和二氢槲皮素均表现出极强清除自由基与DPPH相结合的抗氧化活性,二者浓度分别在0.2 mg/mL时达到92.21%和93.45%,超过BHT;在ABTS实验中,落叶松提取物表现较强的活性,浓度在0.05 mg/mL清除率达到91.43%,二氢槲皮素在浓度为0.2 mg/mL时清除率升高到93.45%,均表现较强的抗氧化活性。而香橙素和圣草酚对DPPH自由基和ABTS自由基的清除活性都表现较弱,均低于50%,抗氧化能力较弱。

     

    Abstract: The main flavonoids and their contents were qualitatively and quantitatively analyzed by HPLC-MS/MS method in the sawdust Larix olgensis var. koreana,which was taken as research object,and their antioxidant activities were also determined. The results showed that three kinds of flavonoids,namely dihydroquercetin(92.07%),aromadendrine(2.39%)and ergophenol(0.19%)were observed in L. olgensis var. koreana. Strong antioxidant activity against DPPH free radical were detected in both L. olgensis var. koreana flavonoids extract and dihydroquercetin,which were higher than BHT and the data reached 92.21% and 93.45% at concentration of 0.2 mg/mL,respectively. The strong activity by L. olgensis var. koreana flavonoids extract against ABTS free radical which reached 91.43% at concentration of 0.05 mg/mL were also observed,even the higher ABTS free radical clearance rate increasing to 93.45% was investigated with dihydroquercetin at concentration of 0.2 mg/mL. The weaker antioxidant capacity were revealed by citrin and eriodictin in DPPH and ABTS experiment,the data were both lower than 50%.
