阿胶蛋白质组学研究

Protein Components of Protein in Colla Corii Asini

  • 摘要: 为探究阿胶药理活性相关蛋白组分,采用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis,SDS-PAGE)和三合一高分辨质谱等技术手段揭示阿胶蛋白质的组成。结果表明,阿胶蛋白在10~250 kDa间呈弥散型分布,通过与马科蛋白图库进行对比,检索出316种可识别的蛋白。研究发现,阿胶含有核心蛋白聚糖、双糖链蛋白聚糖、Lumican、纤调蛋白等富含亮氨酸的小分子蛋白聚糖和免疫球蛋白、胶原蛋白等多种活性蛋白。进一步对阿胶蛋白进行生物信息学的分析,发现阿胶蛋白主要参与系统性红斑狼疮、蛋白质消化吸收、ECM受体相互作用、病毒致癌、肥厚型心肌病、扩张型心肌病等信号通路,这为阿胶药理活性成分的筛选及深层次开发提供了理论基础。

     

    Abstract: To explore the pharmacological activity of donkey hide gelatin related protein components, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and Orbitrap Fusion Lumos Tribrid were used to reveal the composition of Colla Corii Asini (CCA), which would provide a basis for the exploration of protein components related to the pharmacological activity of CCA and further development of CCA. The results showed that the molecular weight was distributed in the range of 10~250 kDa. By comparing with the equine protein library, 316 proteins were retrieved. Results found that CCA contained a variety of active proteins, including leucine-rich proteoglycans, such as decorin, biglycan, Lumican, fibromodulin, and other active substances such as collagen and immunoglobulins. Further bioinformatics analysis of proteins in CCA revealed that these proteins are participate in signal pathways such as systemic lupus erythematosus, protein digestion and absorption, ECM receptor interaction, viral carcinogenesis, hypertrophic cardiomyopathy, and dilated cardiomyopathy. The screening of pharmacologically active ingredients provides a theoretical basis. This results would provide a theoretical basis for the screening and further development of pharmacological active components of CCA.
