香灰菌发酵茶水提物对高血脂小鼠脂质代谢的影响

Effects of Aqueous Extracts from Annulohypoxylon stygium Fermented Tea on Lipid Metabolism in Hypolipidemic Mice

  • 摘要: 为探究香灰菌发酵茶水提物(ASTAE)的降血脂以及肝脏保护功效,采用高脂饲料诱导构建高血脂小鼠模型,将80只昆明小鼠随机分为空白对照组(CK)、模型组(HF)、阳性对照组(UA,200 mg/kg)、绿茶组(GT,400 mg/kg)、香灰菌组(AS,400 mg/kg)以及香灰菌发酵茶水提物高(HA,400 mg/kg)、中(MA,200 mg/kg)、低剂量组(LA,100 mg/kg)。造模成功后,各组小鼠灌胃相应浓度的药物或茶汤,5周后,检测血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及谷丙转氨酶(AST)、谷草转氨酶(ALT)、肌酐(Scr)、尿氮素(BUN)和脂联素(ADPN)的含量;测定脏器指数,通过 HE、油红O染色法观察肝脏脂肪病变及分布情况。结果显示:与 HF 组相比,HA 组能极显著降低高血脂小鼠 TC、TG、LDL-C、AST、ALT、BUN、Scr水平(P<0.01);极显著升高 HDL-C、ADPN 水平(P<0.01);HA、MA 组可明显改善肝脏组织病理学结构,减少肝脏脂肪颗粒。结果表明 ASTAE 可以改善高血脂小鼠血脂异常,减少高血脂症小鼠肝脏中脂质沉积,其机制可能是通过调节高血脂小鼠体内脂联素含量水平,来实现对脂肪代谢的调控作用,进而达到降血脂、改善脂肪肝和保护肝脏的作用。

     

    Abstract: To study the effect of Annulohypoxylon stygium fermented tea aqueous extracts (ASTAE) on blood lipids and liver protection in mice, 80 male KM mice were established hyperlipidemic model by high-fat diet induction, and then randomly divided into eight groups: blank control group (CK), model group (HF), positive control group (UA, 200 mg/kg), green tea group (GT, 400 mg/kg), Annulohypoxylon stygium group (AS, 400 mg/kg), and high (HA, 400 mg/kg), medium (MA, 200 mg/kg) and low (LA, 100 mg/kg) dose Annulohypoxylon stygium fermented tea aqueous extracts groups. After 5 weeks, serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT), serum creatinine (Scr), blood urea nitrogen (BUN) and adiponectin (ADPN) levels were detected. Detect the organ index, and liver fat lesions and distribution were observed by HE and oil red O staining. The results showed that compared with the HF group, the HA group could extremely significantly reduce the levels of TC, TG, LDL-C, AST, ALT, BUN and Scr in hyperlipidemia mice (P<0.01). The levels of HDL-C and ADPN were significantly increased (P<0.01). It could improve the degree of fatty liver histopathology and reduce liver fat particles in the HA and MA groups. The results suggested that the ASTAE could improve dyslipidemia in hyperlipidemia mice and reduced lipid deposition in the liver of hyperlipidemic mice. The mechanism might be by increasing the level of adiponectin and regulating fat metabolism, to reduce blood lipids levels, improve the fatty liver disease and protect liver.
