Optimization of cellulase- assisted extraction of sugarbeet boot pomace crude polysaccharides, compositions and antibacterial activity

  • In order to study the different parameter of crude polysaccharides from sugarbeet boot pomace, the best extracting process was ensured.Based on one- factor- at- a- time experiments, response surface methodology with four- factors- three- levels was used to optimize the cellulase- assisted extraction of crude polysaccharides from sugarbeet boot pomace ( SCPs) . The physicalchemical compositions, monosaccharide compositions and antibacterial activity of SCPs were investigated. The optimal extraction conditions was found as 30 ∶ 1 of distilled water-to-sugarbeet boot, hydrolysis time 40 min, hydrolysis temperature 52℃, p H4.5, and cellulase amount 4677 U / g.Under these conditions, the extraction yield of SCPs was 3.93%. The antibacterial assay showed that these polysaccharides had different antibacterial activity against Staphyloccocus aureus, Bacillus subtilis and Escherichia coli, and the corresponding minimum inhibitory concentrations ( MICs) were 12.5, 25 and 25 mg / m L, respectively.
