Optimization of enzymlysis conditions of geniposide using response surface methodology

  • Objective: This study aimed to optimize the enzymlysis conditions of geniposide. Method: Genipin concentration as response value, the experimental factors including enzyme concentration, enzymlysis temperature, p H and enzymlysis time were investigated to optimize the enzymlysis conditions of geniposide, and the mathematical mode was established by response surface method ( RSM) .Result: The optimum conditions obtained by RSM were as follows, genipin yield was most significantly affected by enzyme concentration, followed by enzymlysis temperature, p H and enzymlysis time.The concentration of β-glycosidase was6.6 m L ( 140 U) , enzymlysis temperature was 56.5 ℃, p H was 4.0, enzymlysis time was 150 min.Under the optimal conditions, the predicted genipin concentration by mathematical model was 4.80 mg/m L, while the experimental concentration was4.66 mg/m L, with a difference of less than 3%. Conclusion: The optimum enzymlysis conditions of geniposide by RSM was convenient and feasible.
