Hypnotic Effects of Ganoderma lucidum Triterpene Acids in Mice

  • Objective:To study effect of Ganoderma lucidum on the sleeping improvement in mice. Methods:50 ICR male mice were divided into 5 groups,which were solvent control group,diazepam group,and Ganoderma lucidum triterpene acids with high-dose group(100 mg/kg),medium-dose group(T50 mg/kg),and low-dose group(T25 mg/kg). Each test group was orally administered 5% tween 80,2.5 mg/kg diazepam and different doses of Ganoderma lucidum triterpenic acid. After 30 days,the effects of Ganoderma lucidum triterpenic acid on the direct sleep test,the extending test with sodium pentobarbital,the hypnosis test under subthreshold dose of sodium pentobarbital,and the incubation period test with barbital sodium were stuides. Results:The Ganoderma lucidum triterpenic acid was administered orally for 30 days,which had no effect on the weight gain of mice. The Ganoderma lucidum triterpene acids had no significantly direct sleep function on mice. The triterpene acids significantly not only prolonged pentobarbital-induced sleeping time at the doses of 25,50,and 100 mg/kg(p<0.01),but also reduced sleep latency at the doses of 50 and 100 mg/kg(p<0.05),compared to solvent control group. In the hypnosis test under subthreshold dose of sodium pentobarbital,triterpene acids at 100 mg/kg significantly stronger than that of diazepam group(2.5 mg/kg),which showed synergistic effect on subthreshold dose hypnosis with sodium pentobarbital. In the barbituric sodium sleep latency test,the sleep latency of Ganoderma lucidum triterpenic acid group was significantly shortened(p<0.01). The high dose at 100 mg/kg had a stronger effect than diazepam group(p<0.01). Conclusion:Ganoderma lucidum triterpenic acid had the function of improving sleep.
